 Synthesis and Biological Evaluation of the 3‐Phenyl‐1,4‐benzothiazine Analogues

Contrasting bacterial resistance to antimicrobial agents by the inhibition of efflux pumps is a promising strategy aimed at restoring antibacterial MIC in resistant strains. In an attempt to improve the benzothiazine class of Staphylococcus aureus NorA inhibitors in terms of efflux inhibitory activity and chemical stability, the synthesis of six different sets of derivatives was performed. The 2-phenylquinoline series provided the best derivatives, suggesting this scaffold as the best suited for further optimization.

Lighting the LAMP: We developed loop-mediated isothermal amplification (LAMP) reactions transduced by oligonucleotide strand displacement (OSD) probes in which competition between defined numbers of “false” (introduced) and “true” (analyte) amplicons leads to order of magnitude analyte quantitation by a single visual endpoint determination. These LAMP-OSD reactions directly and quantitatively estimated numbers of nucleic acids in complex biospecimens.

Call for immunity: A sialyl-Tn (STn) cyclopeptide scaffold bearing the helper T-cell peptide epitope (PADRE) is synthesised. Although the resulting glycoconjugate lacks the native MUC1 peptide sequence, its ability to interact with sialyl-Tn monoclonal antibodies is confirmed in ELISA assays. The availability of sialylated carbohydrate antigens opens up possibilities for the development of a new generation of synthetic antitumour vaccines.

Situation stabilized: Lipid-like peptides (LLPs) can significantly stabilize detergent-solubilized integral membrane proteins in vitro. This stabilizing effect depends on the chemical nature of the LLP and the intrinsic stability of a particular integral membrane protein (IMP) in the detergent. Screening a subset of LLPs is sufficient to stabilize a particular IMP, which can have a substantial impact on the crystallization and quality of the crystal.

Thioether Iron Complexes [(X‐SMe‐7,8‐C2B9H10)Fe(C6H6)] (X = 9 or 10) as Synthons of Neutral Ferracarborane Fragments

The replacement of the benzene ligand of the thioether-substituted ferracarborane [(&#x3B7;-9-SMe-7,8-C2B9H10)Fe(&#x3B7;-C6H6)] by other ligands makes it a convenient synthon of the neutral fragment [Fe(&#x3B7;-9-SMe-7,8-C2B9H10)]. DFT calculations suggest that the lone electron pair of the sulfur atom facilitates these reactions. The structures and electrochemistry of the complexes obtained are also described.

The introduction of oxygen atoms contribute to the crosslinking between carbon layers and adjust the disorder degree of MCMBs by increasing defect sites. The larger interlayer spacing and increased defect sites caused by oxidation are beneficial for MCMBs to store and transfer sodium ions.

Ruthenium(II)-catalyzed C−H bond alkylation of arylphosphine oxide with functional alkenes has been developed. The use of acetic acid was found to be critical to favor the formation of the alkylation products whereas the use of NaHCO3 affords alkenylation products. This method allows the desymmetrization of diarylphosphine oxides. After reduction of the phosphine oxide, novel phosphines bearing a flexible pendent carboxylate were synthesized.

A biocatalytic funnel: The presented concept is a rational approach to tune the concentration of toxic aldehyde intermediates in living cells. We adjust the redox equilibrium between alcohol, aldehyde, and carboxylic acid species by combining an alcohol dehydrogenase with a carboxylic acid reductase to maintain the aldehyde concentration below the toxic levels, yet the reactive aldehyde species are freely available for subsequent aldolase-mediated reaction.

Cellulose was directly converted to C3+C4 hydrocarbons by a bifunctional catalysis of Pt supported on zeolite catalyst, even at 443 K, under hydrothermal condition. Because of their structural sensitivity, Pt/zeolite catalysts with Pt particles larger than 5 nm showed higher TOF values.

Amide-bond cleavage: Zinc-catalyzed esterification of unactivated tertiary amides is achieved with the assistance of intramolecular acyl rearrangement. This reaction can be applied to the one-pot removal of various amide-based directing groups under mild reaction conditions by using an epoxide as an external source of the N-&#x3B2;-hydroxy group.
